Job Description

 

Lumen is seeking a highly motivated, innovative Lead Technician for the Biologics team.


The Laboratory Technician will work closely with and support the scientists on the Biologics research team.  The work will involve several projects aimed at developing biologic therapeutics using Lumen’s Spirulina-based expression system.  The individual will be expected to perform a wide range of molecular biology techniques, techniques in bacterial protein expression and purification, and biochemical and biophysical techniques aimed at investigating protein folding and protein-protein interactions.  The position requires the ability to work as a team member and to assist other laboratory personnel as needed.  The position is anticipated to be central to Lumen’s acquisition and integration of new and complex technologies, and to sharing such capabilities with other company teams, and as such, flexibility and independence will be essential.  The position requires strong organizational skills and meticulous laboratory record keeping. Effective communication skills are crucial in coordinating with Lumen’s research and production scientists, in providing status updates, and in raising quality or troubleshooting issues as they may arise.

 

Essential Duties and Responsibilities

 

The major tasks for this position are as follows:

 

  • Antibody engineering

    • Tasks include: protein design/modeling, SnapGene software use, SDS-PAGE, ELISA, in vitro adherence assay, HI assay, aseptic Spirulina culture.

  • Stability analysis of novel antibodies

    • Tasks include: Enzymatic digestion, preparation of GI-tract simulating buffer and enzyme conditions, SDS-PAGE, silver staining, ELISA, thermal shift assay, circular dichroism and others TBD.

  • Bacterial Protein Expression

    • Tasks include: expression vector cloning, culture growth and induction, analytic and preparative chromatography (SDS-PAGE, SEC, affinity chromatography); protein modification (biotinylation, covalent assembly, enzyme-mediated cleavage).

  • In vitro antibody functional screening

    • Tasks include: mammalian cell culture, mammalian RBC handling, virulent bacterial cell culture (including quantitative), aseptic technique, qPCR.
       

Desired Qualifications & Requirements

 

Education & Experience

  • Possession of BA/BS degree in biochemistry, cell biology, biotechnology or related field of study 

  • Minimum of five years’ experience in protein sciences or related area. 

    • Experience in bacterial protein expression techniques, affinity-based protein purification.

    • Experience in performing cell based and biochemical /immunochemical assays (ELISA, SDS-PAGE, Western Blot, etc.).

    • General molecular biology skills including cloning, DNA isolation, PCR and Q-PCR.

    • Experience with size exclusion chromatography preferred.

    • Previous experience performing expression and purification of large quantity proteins preferred.

    • Previous experience characterizing protein-protein interaction preferred.

Physical Requirements:

 

  • Sit for extended periods of time (2 or more hours)

  • Occasionally lift or carry up to 20 kg

Lumen Bioscience was founded in early 2017 to make and commercialize ultra-low-cost biologics, such as antibodies, in our proprietary spirulina expression platform. We design and manufacture therapeutics against disease targets that have been inaccessible to the pharmaceutical industry because of the astonishingly high cost of using traditional methods to make and deliver biologic drugs. Now, for the first time, Lumen can make antibodies and other biologics at a cost that allows for daily, high-dose, oral delivery.


Our current programs focus on diseases that can be prevented or treated by an orally delivered therapeutic; the target markets are enormous, the treatment modes of action are well-researched, and the unmet medical need is obvious. We have been fortunate to have earned financial support for these programs from groups like the Bill & Melinda Gates Foundation and the NIH.  In the longer term, we believe that our unique technology–––the ability to safely deliver frequent, large doses of a wide range of biologic compounds–––will become commonplace in biopharma as the science continues to advance and uncover new opportunities in the fields of microbiome signaling, metabolism, inflammation, and oral delivery of systemic biologics.
